Protein spots are chemically modified and ultimately cleaved using
trypsin an enzyme which cleaves proteins at specific amino acid
residues in the polypeptide chain. This generates a mixture of peptides
whose masses can act as a “fingerprint” of the parent

Voyager DE PRO MALDI tof mass spectrometer
The MALDI tof mass spectrometer measures the masses of the tryptic
peptides generated by the MassPrep station. Matching these peptide
masses against a database provides protein matches which using bioinformatic
packages can provide information about the protein of interest in
a particular biological process. Further information about proteins
which cannot be identified by MALDItof can be obtained by submission
of the tryptic peptides to nanoflow HPLC and ESI mass spectrometry
using the ABI Q Trap. |
